I put a nice thick ribbon of plumbers putty around the drain flange as wide and as thick if not thicker than the drain flange (the flange is usually about 1/2". That way, I an SURE that I have plenty of putty to stop leaks later when I tighten it down to the sink.
As to the 4 holes, I think you're talking about the drain holes in the drain tailpiece? If so then that is for the overflow drain of the sink, to allow the overflow water to get back into the drain.
