My wife and I are priming to cover up smoke smells and nicotine stains but arent sure which is the better choice. I understand O. Kilz is oil based and 2 is latex. So if I use O. Kilz, do I then need to prime with something else before I cover with a latex paint?

You can put the latex over either Kilz. I would wash the walls well with Dirtex before priming.
