I live in Puget Sound Washington and we’re replacing cedar shingles on the south side of our 1914 house that were not well kept by previous owners. Is it worth upgrading to pre-primed shingles (primed on all 6 sides)? I’ve been quoted twice the price for pre-primed and wonder if it is worth it. Also, if we go with pre-primed, how long after installation should they be painted?

How much are you talking? If the unprimed shingles are $1,000 and the primed are $2,000 I say get the primed. If you're talking $5,000 and $10,000 I'd say "no."
They are generally worth it. The shingles will be sealed from moisture intrusion. Think about how much work it would be to prime all six sides?
Got the time to dip prime them yourself? Have the primed shingles painted ASAP. Paint primer usually specifies to top coat within 3 weeks.....don't want to have to top coat dirty primer coat.
That would depend on how much more the primed shingles are. The OP says it's for one wall but doesn't say how many square feet. For $5k difference I can dip a whole lot of shingles in one day and save a ton of money.
