I want to have vinyl installed in place of or on top of my existing masonite or fiberboard siding that is showing neglect from the previous owner. What is the best method tear off masonite and start from sheathign or apply vinyl over the existing siding, if over existing what’s the best method?

tear it off to see what's going on under there....
or fan-fold foam insulation to smooth things out before ya vinyl.
For a bit of real insulation....if deepening the windows/doors into the building don't bother ya....try dow board(blue board)....then inch or so thick stuff.

I like the idea of an additional 1" of insulation, the house is in need of more insulation, the card stapled to the rafters in the attic states the R-values, but I don't believe that those values were actually achieved.
Increasing the trim depth at openings, actually appeals to be as I prefer the look of a thicker wall construction anyway.
