will a box made of standard 2×6 stay straight. my plan is to use a 2×6 constructed box about 60 inches tall cover it in stone tile and used as a newel post

Probably not the best way to do a tile column. I would make it out of 3/4" or 1-1/8" plywood instead, biscuits and screws for the joints, then wrap it in tile backer board using backer board screws.
Depends on what the 2x6 species is.

You might also consider building your post column of concrete half-blocks on a concrete footing.
Place a couple #4 bars in the footing, slip the block over them, mortar the joints, fill the cores.
May sound like a bit of work, but it really won't be and it's a very simple masonry/block-laying project that will give you an excellent surface for your tile.
If framing lumber hasn't had time to dry it's going to shrink a great deal--and twist, and cup, etc. Not good things to have happen under tile.
Plywood gets my vote.
