I need to replace about a 2 ft section of 2X4 sill on a concrete block foundation. Once I replace the section of sill, how should I anchor it to the block foundation? Is it necessary to anchor it?

if the blocks are filled with concrete, use some tapcon.
And if not (I'm not sure), just screw them into the block itself? Should I worry about cracking?

Once you remove that old sill you will know if the cells were filled in the block.
If the section you remove is strapped to help hold down the sill and the structure or already has embedded bolts you should reattach what is already there. I don't know what the structural requirements are in your area but for such a short repair a couple of toe nails in each end to the remaining sill and the toenails in the studs should do it.
