For laying concrete blocks on an uneven slab, can a thicker bed of mortar be used to keep the top of the blocks level? Or will the block simple sink in? I have a old slab that dips for about an 1 inch or so and then comes back up a few feet later.

Yes. That is how it's done. Mix it up stiff for the bed joints of the first course. For differences of more than about 1.5", split bricks are used in conjunction with the mortar.
