My basement is dry and has concrete floor and walls. I’d like to put DriCore or Sub-Flor on the floor and build stud walls inside the concrete block. My question is: Do I put the floor down first and build the walls on top of it, or should I build the walls and then the floor inside the walls?

Put the floor down first, and then build the walls on top of it. You also may want to look at another option, depending on how much of a DIYer you are. Lowes now sells Delta FL, which used to only be available from certain contractor supply places. It's a dimple sheet that comes in rolls. You put a T&G subfloor on top of it which has to be screwed through to the concrete with tapcons or similiar. Don't even think of trying to drill all the holes without a real hammer drill.
It's much faster than Dricore or Subfloor, and cheaper to boot.
