I have a concrete block basement with 2×10 joists. It was built without any provision for drier venting (actually, no provision for laundry at all; another story.) Will structural integrity be compromised if I cut a 4in hole in the centre of the rim joist to accommodate a vent?

Cut the hole. If you use a big Milwaukee drill with a 4 in. hole saw, hang on tight.

None of this matters in geological time.
Go ahead and cut...but make the hole 4-1/8" or you'll have trouble getting the vent hood thru. If the basement is unheated (or even cool) slope the duct to drain any moisture to the outside.
