Question – I’m building a garage w/2 courses of block above grade. Need to tell mason how big the block opening should be. 84’s garage door salesman says 2″ bigger than door, or 16′-2″. I’ll add 3″ or 3 1/4″ to that to provide a PT nailer on the inside of the block opening. But should I provide more? I haven’t done a garage door in years, but because of the way the roller axles attach to the track – shouldn’t there be more than 1 1/2 inches on either side?
Also, G-P’s laminated headers are sized for a clear span of 16-3. Does this mean the RO should be 16-3 to wood, or 16-6 for the block?

Since you are going to put PT at your nailer(I'll assume 2x material) Have the mason make the block opening 3 inches bigger than the door. Then when you hang the door, you'll only loose the stop thickness from the width, and it will cover the edges of the door without rubbing the finish off the door.
*Im with Keith if the garage door people put stop or weatherstripping on the jambs. If not the salesman probably has the right idea. We usually frame for a finished opening 1" less than the specified door width as the salesman recommend. Most of the garage door companies down here dont install the weatherstripping/stop unless there is a crack to fill. If your jamb material is something other than 2x material then adjust accordingly. Also be dang sure both jambs are plumb cause there is little room for adjustment.
*I'm jumping ahead in your timeline a bit, but before you have your overhead doors installed, get your trim carpenter to finish the inside opening with some nice 1 x 6's. Paint or stain before the hardware is attached. Makes the cleanest appearance. Of course, I'm assuming you're going to drywall the inside.

I'm going to make the rough wood opening 16-0, and 1X trim will give me some space on each side to hide the gaps from my out of plumb jambs.I've since learned that Clopay requires 3 3/4 " on each side for installing the track. Since I'd rather not space the door back the thickness of the block, I'll close in the opening in the block to 16-0 rough, or 15-10 1/2 finished.
