I have made some wainscotting using MDF and I’d like to paint them…Should I use a sealer first, then prime?
What would you suggest?

Forget the primal scream, just ROAR!!!
I would use a primer/sealer like one of the products that Zinser sells. For any exposed ends, you can use spackling or drywall mud to get an even look. The ends are pretty porous so prime it really good or spackle it. Prime and paint it.

No sides will be exposed...No it is not pre-primed...
The only problem in painting MDF comes with the exposed edges. Since you have none, you can just prime and paint as you would any regular wood surface. I would suggest you sand the primer.********************************************************

What Nikki said. Oil primer, because oil penetrates, then sand the primer and maybe even reprime.

I agree that an oil primer is best. BM has a great one called "Alkyd Enamel Underbody"; you can overcoat it with either alkyd or latex paint.********************************************************
