Installing 1×6 v-groove siding in an attic remodel. Client wanted a clear finish, so before installing the wood I lightly sanded then put on one coat of Minwax water based polyurethane. It obviously needed another coat, but it was a start. Half way through the clients DW decided it should be painted white. I wnet ahead and applied one coat of poly to all the rest of the boards before they were installed so they would all be starting from the same point before painting.
For whatever reason, the client bought a gallon of Porter interior latex in their favorite color (white). What do I need to do to the poly before applying the latex? It has cured about 4-5 days now.

Sand it well with 100 grit. I would use a quality oil base primer, sand that lightly then apply your paint.
pretty much same I would do. That first sanding is to rough up the poly surface to give the new primer a mechanical bond. After that, the second light sanding would be with 120 grit to smooth things out between coats. Maybe 180 grit before final coat.

Well, the first (and only) coat of poly was a bit thin, and it tended to raise the grain, leaving a rough surface and uneven coat. The first coat of paint on a sample section was also a little thin, and the HO likes that because it allows some of the grain to show through ... like a semitransparent stain or old weathered paint.
I'm sorry, I thought you wanted it done the right way.
