I want to span one joist space in a floor, 16 inches wide by 3 feet long, below a skylight to allow some light to go through the floor into the room on the story below. It will be stepped on only by socks and bare feet so scratching isn’t an issue. What thickness of plexiglass or lexan do I need to support people stepping on it without giving way? Plexiglass or Lexan?

Lexan. Much stronger than plexi, won't shatter, much more scratch resistant. You probably need to talk to a supplier, though, to find out what thickness is appropriate.
Soxs will scratch plexi , polycarbonate will be much more scratch resistant. It is also a lot stiffer than plexi.
I can't recomend a thickness ,but I have a 112"x12" x3/8" polycarb sheet that is almost impossable to bend. Maybe 1/2 or 3/4 would do . This recomendation is worth every penny that you paid for it.
My primeval analysis suggests that this room must be a bathroom since where else would only socks and barefeet be used?
I would suggest 1" thick plate glass for best visibility.

Plexiglass is a brand of acrylic. Lexan is a brand of polycarbonate. Definitely polycarbonate. You can buy polycarbonate treated to be more scratch resistant.
Of course, an option is to go with something patterned or frosted, or apply frosted contact paper to the underside like the kind used on bathroom windows when the neighbors are too close.
