Hi, I’m a newby here, and have been woodworking since 1986 in a 20 x 30 shop with a concrete floor. I’ve recently moved and am designing a new shop. I’m limited to 16 wide x 45 long and 16 high. I have ordered a steel building that size.
I’d like NOT to have a concrete floor and wonder if I can: put down a vapor barrier, treated 2 x 4, 1″ blue board, treated 3/4 plywood, and cover it with 3/4 T & G pine floring availably locally from a mill.

A little insight as to where you are and yur climatic conditions would help... What is going to setting on the floor? Is the shop hated? Is drainage a problem or will things stay dry under ant conditions? Helps to put a little info in yur profile too...
Why do you think you need the pt ply????
How about VP, gravel or sand, 2x6 joist, blue board in the bays just under the floor deck w/ a 1/2" air space, 1/2 or 5/8" ply sub floor and then yur T&G??? Spanning 16' is gonna be tough for 2x6's unless you put in intermediate support say at center line or at 1/3 interval.
12"OC the 2x6's and skip the ply???
If I read yur idea correctly - level woud be hard to achive, the floor would have way much flex to it and dead load ratings would be terrible...

Are you wanting to put a wood floor over CC???
If so..
VB. 8 mil or felt.
2X4 sleepers laid flat... Shoot or tapcon them down. Shouldn't need these in PT because the VB breaks the wood / CC contact.
1-1/2" Blue board to fill the bays.
1/2 or 5/8" cdx.
