This is in new construction. Fireplace insert, dry wall, oak flooring over plywood subfloor (framing the footprint for the hearth), all are in. I plan on using one piece of cut stone for the hearth, about one inch thick. What do I need to use (if anything) under this stone for subfloor protection.

Can't say without seeing the specs on the stoveinsert and the local code restrictions.

I would felt paper at least #30, metal screed held down with hot dip roofers not penetrating sub floor,3/8" min. latex medium bed mortar let set a day or until cured, then comb on thinset mortar with 1/2"x1/2" notch trowel and set stone but like piffin said check specs & codes first.
