In the process of tiling a small half bath on the first floor with a basement below. Currently there is 1x running and plan to use 1/2 ply with 1/4 hardi. Do I need a moisture barrier between 1x and new ply?

You do need to follow Hardi's instructions. That means unmodified thinset below the hardi; nail Hardi to plywood not joists; leave a sixteeth space between Hardi. Tape and mud seams.

putting moisture bar between two plies of wood like that sounds like an invitation for trouble. If I were afraid of leaks coming through, I would put the membrane over the ply and under the Hardi, but then how do you stick the hardi down?
If this is a shower, you need a PVC or other membrane under mud coat but for plain old floor, I'd skipp it.
Are you trying to stop moisture from migrating up or leaking down? That tells you where yopu would want a wet stop
