We will soon be installing prefinished, 5/16″ real-wood oak flooring. Can I nail this to OSB? Can I glue it to OSB? Why would I choose nailing over gluing or vice-versa? If I can’t nail directly to OSB, how thick does the underlayment-plywood need to be?

I sure wouldn't glue to OSB. I'd worry about it delaminating after a few seasons of expansion / contraction. I'd staple or nail it down assuming you have a 3/4" subfloor. If not build it up with plywood which is not as likely to blow out when nailed through.

*Let's see... how do I say this tactfully...Since you're using 5/16", I'm guessing that it is of the "pre-finished" variety. Have you considered contacting the manufacturer of the flooring product to see what they recommened for your situation?Often, the waranty of the product is dependent upon following prescribed installation instructions for particular substrates.I'm with David on this one. Also, since we generally put down felt under hardwood floors before nailing them, the glue idea seems inappropriate. Why do we use felt? "Because that's the way its always been done."As for floating the floor, an alternative Daniel offered, I'm of the impression that this is more often used for edge-glued laminate floors.Wish I could be of more help, Steve
*L., I'm in agreement with Steve's Post #4 in regards to contacting the manufacturer for their recommendations. Ask the manufacturer of the flooring if it can be glued. If so, what type of glue. Some require proprietary glues for warranty work, some restrict certain brands from use with their products. From there, ask if that glue can be used over OSB. And, if they weren't supplied, get tech sheets. Always have the paper to back you up, especially if this is someone else's house.Be sure to tell them you're comtemplating 3/8ths hardwood over 3/4" OSB...personally, I consider the underlayment to be a bit light.
*I believe the felt is meant as a vapor retarder. Assuming this is over a crawlspace, moisture from below can wreck havoc with your hardwood. Why not use a full 3/4" hardwood. I think nailing it to OSB would be a mistake -- I just don't think the nails are going to hold long-term. When you start with a sow's ear, you're not going to get a silk purse.
