I am relocating an old door and jam along with a section of wall in an older house. When I build the wall and install the jam and door in the new location (covered with a good hardwood floor) can I screw/nail the bottom and top plate to the existing hard wood floor and wet plaster ceiling? My other option is to channel the floor and ceiling for the plates.

Certainly you can do that on the floor. It may or may not be the best way to handle the ceiling -- you should at least attempt to fasten to joists above if there are any appropriately placed.
Steel studs.
This is a perfect use for steel studs. Screw through the floor and through the ceiling into the joists if possible.
