My shed roof has 2×4 rafters making a 6′ span. I am sistering additional 2×4’s to each rafter. I will use 10d common nails. Would there be any benefit to using a waterproof glue?

I doubt that there would be good enough contact for the glue to be really effective. But it won't hurt anything.
