Size of the unit is determined by the heat loss calculations for the home and the location. Return air vent size is in turn based on the unit size, or more precisely the output cfm.
There is a lot more to answering a question like that than can be easily explained or quessed at. I would suggest you contact a competant HVAC contractor for you specific design needs.
You might get a few ROT guesses here, but most don't like to give them.
