Is it ok to use a single board for poplar (paint grade) slab drawer fronts that are 5 – 5.5 inches wide and 24 inches long? The width is that of a 1×6 but I’m not sure if it would be more stable to glue up 1×4’s and cut to size?

Check your boards for cupping and warping. Precut them (with a few extra) close to your final dimension, then let them sit inside for a week. See if any boards moved and discard them, use the good ones.
