I am replumbing (DIY) a vacation house and need advice on plumbing vent sizes. I am running 2 bathrooms back-to-back each with a toilet, sink, and tub/shower running to a 4 inch line. How large should the vent pipe be?

Round here you need 1 3" main stack in the house and minimum 3" at roof penetrations. A 2" vent would suffice if there is a 3" elswhere in the house.
Different locales may vary.
the totel vent cross sectional should equel your main drain cross sectional
Try the local plumbing inspector. I think the greater concern would be how far fixtures are from the stack, you may need to add a couple Studer valves for secondary venting.
